Buffer containing 50 µM FITC-Hst-5 and 5 µg/ml PI was added to the cells, and uptake of fluorescence was followed by wide-field time-lapse microscopy. The three-dimensional image was acquired from deconvolution using fifteen 0.2 micron optical sections. |
