English: Additional file 1: Movie 1 Visualisation of active Rac in Wild Type cells. AX3 cells expressing the active Rac marker CRIB-GFP (vector pDM841) were induced to develop by starving them for 4 hours and pulsing them for 2 hours with 100 μM cAMP pulses every 6 minutes. Cells were transferred to a glass-bottom dish and overlayed with a thin layer of 0.4% agarose in development buffer. Fluorescence images (green) were collected using TIRF illumination at 1 frame per 2 seconds. The brightfield image is shown in dark blue. The field of view is 83x83 micrometers. |
