Description
English: Supplementary Movie 1 Endothelial RhoA is locally and transiently activated during neutrophil extravasation not during adhesion. Time-lapse recordings of the DORA RhoA biosensor showing spatiotemporal RhoA activation during neutrophil TEM under physiological flow conditions (0.8dyne per cm2). Left panels from top to bottom show, Venus/Cer3 ratio images of the DORA RhoA biosensor, Merge and DIC, respectively. Right panels show zoom of the ROI shown in left panels. Calibration bar shows RhoA activation (Red) relative to basal RhoA activity (Blue). Images were simultaneously acquired by time-lapse wide-field microscopy (Zeiss Observer Z1) using a 40x 1.3 NA oil immersion objective. Frames were taken every 4 s for 6 minutes.
