English: Modern confocal microscopes, built according to the classical scheme, have a resolution of no better than 180 nm in the lateral direction and 500 nm in the axial direction. This limitation does not allow the resolution of many subcellular structures. This has led to different methods to improve or bypass the diffraction limit in optical microscopes. Expansion microscopy (ExM) is a recent addition to the array of optical imaging protocols and is based on the physical expansion of the tissue embedded in the polyacrylate gel matrix (Chen et al., 2015). I used a protein-retention ExM protocol, where 4-4.5x tissue expansion can be performed in one step (Asano et al., 2018), which corresponds to around 100x expansion by volume.
You look at the expanded node of Ranvier (NoR) in the axon of the principal neuron of murine Medial Nucleus of the Trapezoid Body (MNTB). MNTB is one of the centres of sound processing responsible for localization and encoding time characteristics of complex sounds. NoRs are microscopic gaps between Schwann cells in myelinated axons. Their function is to speed up the propagation of action potentials along the axon, maintained by several voltage-gated potassium channels, including Kv3.1 (yellow, anti-Kv3.1b + AlexaFluor 488, laser 488 nm). Expression of the Kv3 channels enables action potentials to be of short-duration and high firing frequency, which renders these channels critical for correct sound processing in the brain. Prep: frozen 12 µm section from CBA WT mouse, P20, expanded using proExM protocol. This image was acquired from a z-stack obtained using Zeiss LSM 980 confocal with a 63x objective. Це обмеження не дозволяє розділити багато субклітинних структур. Це призвело до розробки методів покращення або обходу межі дифракції в оптичних мікроскопах. Розширювальна мікроскопія (ExM) є нещодавнім доповненням до протоколів оптичної візуалізації та базується на фізичному розширенні тканини, вбудованої в матрицю поліакрилатного гелю (Chen та ін., 2015). Я використовувала протокол proExM, де за один крок можна виконати 4-4,5-кратне збільшення біологічного зразка (Asano та ін., 2018), що відповідає приблизно 100-кратному об’ємному розширенню.
Ви дивитесь на розширений вузол Ранв’є в аксоні головного нейрона медіального ядра трапецієподібного тіла миші (MNTB). MNTB є одним із центрів обробки звуку, що відповідає за локалізацію та кодування часових характеристик складних звуків. Вузли Ранв’є - це мікроскопічні проміжки між шванівськими клітинами в мієлінізованих аксонах. Їх функція полягає в прискоренні поширення потенціалів дії вздовж аксона, що підтримується кількома потенцiалзалежними калієвими каналами, включаючи Kv3.1 (жовтий, анти-Kv3.1b + AlexaFluor 488, лазер 488 нм). Експресія каналів Kv3 дозволяє потенціалам дії бути короткочасними та високочастотними, що робить ці канали критичними для правильної доставки та обробки звуку в мозку. Підготовка: заморожений коронарний зріз (12 мкм) мозку миші CBA WT (дикий тип), P20, розширений за протоколом proExM. Це зображення було отримано з z-стека, отриманого за допомогою Zeiss LSM 980 confocal з об’єктивом 63x.
