Description
English: Supplementary Movie 1 Time-lapse of flagellum disassembly following promastigote uptake by a macrophage. Example confocal epifluorescence time-lapse videomicrograph of promastigote uptake by a BMDM. BMDMs were pre-incubated with 10,000 Mw rhodamine-conjugated dextran for 18 h prior to infection, then co-incubated with SPPs expressing SMP1::eGFP at a target MOI of 3, at 34°C with 5% CO2 in the environmental chamber of the microscope. Images were captured every 5 min. Instances of parasite death are visible through increased cytoplasmic dextran signal. Stills from this videomicrograph are shown shown in Fig. 2a. Scale bar represents 10 μm.
