English: Movie S1. Effect of NSC23766 on human plaque-induced platelet thrombus formation under arterial flow conditions. Hirudin-anticoagulated blood was incubated with mepacrine to visualize platelets by fluorescence. Blood was perfused (direction right to left) over atherosclerotic plaque-coated microfluidic chambers and observed for 10 min. Upper channel, control; lower channel, blood pre-treated with 300 μM NSC23766. In the upper channel, rapid platelet adhesion and aggregate formation (green fluorescence) occurred, mainly at the edges of the channel, where also the majority of plaque material is present (as seen by phase contrast microscopy before start of the flow experiments). NSC23766 reduced platelet adhesion and aggregate formation. The video is in. mov format and can be viewed using Quick time player on different PCs with Windows XP or Vista. |
