Description
English: Photobleaching enables single particle tracking of GFP-Sec4p at the cell cortex. Time-lapse videos of two representative wild-type cells (BY4741) expressing both GFP-Sec4p and Abp1p-RFP. Large white circles indicate the photobleached region (bud membrane), and asterisks indicate the duration of the laser burst. Green arrowheads indicate cortical GFP-Sec4p particles, RFP-Abp1p is indicated by red arrowheads, and yellow arrowheads indicate the transitional overlap of GFP-Sec4p and RFP-Abp1p fluorescence. Total acquisition time is 90 s edited and compressed to play at 5 frames/s (for technical reasons, 3 frames during laser photobleaching were excluded).
