English: A single embryo of the fruit fly Drosophila melanogaster imaged from four different angles during the last two syncytial cell divisions and early gastrulation movements.
I collected embryos from transgenic flies expressing a fluorescent membrane fusion-protein (Gap43myr::mCherry from Martin et al. 2010) and embedded them in low melting agarose inside a glass capillary, as described in the OpenSPIM sample preparation. I mounted the sample in a Zeiss Lightsheet Z.1 microscope and acquired four different angles of the same embryo, each view containing a stack of ~200 slices, every 90s. Using Fiji, I improved the signal-to-noise ratio by processing the slices through a Median filter (radius=1px) > Smooth filter > Subtract Background (radius=20px), and by running a custom macro, I created a maximum intensity projection for every timepoint using the Temporal-Color Code plugin with the mpl-viridis LUT to color code the Z-depth. Finally, I arranged the four views using the non-linear video editor Kdenlive to show from left to right: the right side, back/dorsal, front/ventral and left side of the embryo, respectively.
