Movie 2, PIP2 dynamics associated with an expanding actin wave . The cell expressed PH-PLCδ1-GFP as a label for PI(4,5)P2 and mRFP-LimEΔ for filamentous actin. During propagation of the wave, the PIP2 label declined while the external area in front of the propagating wave was converted into internal territory. The left panel shows fluorescence in the mRFP channel, the middle panel simultaneously recorded fluorescence in the GFP channel. The right panel displays fluorescence intensities of PH-PLCδ1-GFP (green) and mRFP-LimEΔ (red) along the line scan superimposed on the images in the first frame. Frame-to-frame interval 1 s. |
