English: Montage of a two-color time-lapse of Klp61F-depleted cells. The RepEg5 S2 cell expressing GFP-α-tubulin (upper left; green in merge) and human Eg5-mCherry (upper right, red in merge) is next to a cell that is not expressing Eg5-mCherry. Note that at the beginning of the movie the RepEg5 cell has a bipolar spindle while the nearby cell that is not expressing Eg5 has a monopolar spindle. The bipolar spindle in the RepEg5 cell rapidly collapses into a monopole following the addition of 10 μM STLC between frames 00:00 and 02:00 and remains a monopole for the duration of the experiment. Following addition of STLC, spindle-associated levels of Eg5-mCherry drops while cytoplasmic mCherry fluorescence increases. Lower left panel is the merged time-lapse. Scale bar is 10 μm. |
