[edit]DescriptionChlorophyll A autofluorescence in chloroplasts separated by spectral imaging from the FM 4-64 dye signal in plasma membranes of pavement and stomatal cells of Arabidopsis cotyledons.tif |
English: Сhlorophyll A autofluorescence (685–740 nm, marked red) in living plant tissues makes a detection challenge for confocal microscopy in terms of separation of several signals because of the overlap of the emission detected from several fluorophores. The styryl dye FM 4-64 ((N-(3-triethylammoniumpropyl)-4-(6-(4-(diethylamino) phenyl) hexatrienyl) pyridinium dibromide) selectively stains membranes with magenta fluorescence (excitation/emission maxima ~515/640 nm). To separate these two signals, lambda mode with further linear unmixing and online fingerprinting approach in Zeiss Zen 2012 software. Image was documented with a Zeiss 710 confocal laser scanning microsope (CLSM) platform (Carl Zeiss, Germany) at the Centre of the Region Haná for Biotechnological and Agricultural Research, (Olomouc, Czech Republic) by the research fellow of the "Institute of Food Biotechnology and Genomics of National Academy of Sciences of Ukraine" (Kyiv, Ukraine). Laser line used for FM 4-64 and chlorophyll A exitation is 514 nm. Scale bar: 10 um.
