English: Сhlorophyll A autofluorescence (685–740 nm, marked red) in living plant tissues makes a detection challenge for confocal microscopy in terms of separation of several signals because of the overlap of the emission detected from several fluorophores. Green fluorescent protein (GFP)(marked green) with the excitation/emission maxima ~488/509 nm is now routinely used as a tag to different intramembrane proteins as in case of the transgenic Arabidopsis line on the image. To separate these two signals, lambda mode with further linear unmixing and online fingerprinting approaches have been used in Zeiss Zen 2012 software. Image was documented with a Zeiss 710 confocal laser scanning microsope (CLSM) platform (Carl Zeiss, Germany) at the Centre of the Region Haná for Biotechnological and Agricultural Research, (Olomouc, Czech Republic) by the research fellow of the "Institute of Food Biotechnology and Genomics of National Academy of Sciences of Ukraine" (Kyiv, Ukraine). Laser line used for GFP exitation is 488 nm. Scale bar: 10 um. |

Автофлуоресценція хлорофілу А у хлоропластах, відділена від сигналу зеленого флуоресцентного білка (GFP) у плазматичній мембрані за допомогою спектрального отримання зображень. Як відомо, автофлуоресценція хлорофілу A (685–740 нм, позначена червоним) значно перешкоджає детекції емісії різноманітних флюорофорів при конфокальній мікроскопії живих рослинних мікропрепаратів. GFP (максимуми збудження/випромінювання ~488/509 нм, позначений зеленим) використовується для маркування різноманітних внутрішньомембранних білків, як у випадку трансгенного арабідопсиса на представленому зображенні. Для розділення двох сигналів використано функції Lambda mode, Linear unmixing та Оnline fingerprinting у програмному забезпеченні Zeiss Zen 2012. Зображення отримано за допомогою лазерного скануючого конфокального мікроскопа Zeiss 710 (Carl Zeiss, Germany) у Центрі біотехнологічних та сільськогосподарських досліджень регіону Хана (м. Оломоуц, Чеська Республіка) науковим співробітником ДУ ”Інститут харчової біотехнології та геноміки НАН України” (м. Київ, Україна). Для збудження емісії GFP використано лазер з довжиною хвилі 488 нм. Масштаб мікрофотографії: 10 мкм.
