Description
English: Supplementary Movie 7 Tracking of CD3ζ-bearing vesicles at the IS in control Jurkat cells. Control Jurkat T cells transfected with CD3ζ-mCherry were allowed to settle on stimulatory anti-CD3/CD28-coated surfaces and recorded under a TIRFm, at a 200 nm of penetrance upon excitation with a 561 nm laser. Images were taken every 100 ms. MLN8237 or vehicle was present in the imaging medium. Imaris Software was used to recognize fluorescence corresponding to the vesicles and to calculate the trajectories, their duration and the speed of the vesicles. Movie was mounted at 20 fps.
