English: 4D two-photon time-lapse imaging enables direct observation of radial glial cell migration. GFP-expressing cells are shown after transfection with BLBP-GFP plasmid at E14. A single timepoint (125 h after electroporation) is shown initially with high spatial resolution. As the time-lapse imaging proceeds (one complete z-stack acquired every ~30 min), GFP-expressing cells can be seen migrating out toward the pial surface using somal translocation over a time period of 10 h (out of the 168 h total imaging time) at a high spatial and temporal resolution. In this sequence dorsal is toward the top and the pial surface is toward the right of the frame. Single grid square is 20 × 20 μm. |
