English: Isolation of gametangia (antheridium and archegonium) and respective paraphyses of the species Mnium punctatum
The species Rhizomnium punctatum or Mnium punctatum is part of the group of foliose mosses. Rhizoids can be compared to root hairs, cauloid to the stem, while phylloids to the leaves of better developed plants. The gametophyte reproduces sexually and forms sexual organs - gametangia, which are usually located at the top of the cauloid, protected by top phylloids. Between the gametangia there are paraphyses whose function is to maintain humidity. Between the male sex structures (antheridia) there are paraphyses whose top cells are spherically expanded. On the other hand, the cells that build the paraphyses, which are located between the female sex organs (archegonium), are of the same shape. The male and female sex structures are formed on separate individuals, which is why this species is a dioecious plant. For the preparation of a native microscope slide and isolation of the gametangia with appropriate paraphyses, the following is needed: object and cover glass, a water dropper, tweezers and a sample of the plant material. On the object glass, with the water dropper, a larger drop of water is dropped centrally and a smaller drop of water is placed peripherally. With the tweezers, the phylloids are removed in order to free the top part of the cauloid - the place where the sex structures and paraphyses are located. The released tip of the calluloid is placed in the central drop of water on the object glass and macerated using tweezers. The gametangia and paraphyses break off and are released into the drop of water. Excess plant material is removed from the drop of water. With a cover glass, placed at an angle of 45°, the peripheral drop of water is stretched towards the central one. After joining them, the cover glass is lowered onto the object glass. Видот Rhizomnium punctatum или Mnium punctatum е дел од групата на фолиозни мовови за кои карактеристично е тоа што гаметофитот е изграден од ризоиди, каулоид и филоиди. Ризоидите може да се споредат со кореновите влакненца, каулоидот со стебленцето, додека филоидите со листовите на подобро развиените виши растенија. Гаметофитот се размножува полово и формира полови органи – гаметангиуми кои најчесто се сместени на врвот од каулоидот, заштитени со врвни филоиди. Помеѓу гаметагиумите се наоѓаат парафизи чија функција е одржување на влажноста. Помеѓу машките полови структури (антеридиуми) се наоѓаат парафизи чии врвни клетки се топчесто проширени. Од друга страна, клетките кои ги градат парафизите кои се наоѓаат меѓу женските полови органи (архегониуми), се со ист облик. Машките и женските полови структури се образуваат на посебни единки поради што овој вид е дводомно растение. За подготовка на нативен препарат и изолација на гаметангиуми со соодветни парафизи потребно е: предметно и покровно стакленце, капалка за вода, пинцета и примерок од растителниот материјал. На предметно стакленце, со капалката за вода се капнува, средишно, една поголема капка вода и странично, помала капка вода. Со помош на пинцета се отстрануваат врните филоиди со цел да се ослободи врвниот дел од каулоидот - место каде што се сместени половите структури и парафизите. Ослободениот врвен дел на калулоидот се поставува во средишната капка вода на предметното стакленце и се мацерира со помош на пинцета. Гаметангиумите и парафизите се откинуваат и ослободуваат во капката вода. Вишокот растителен материјал се отстранува од капката вода. Со покровно стакленце, поставено под агол од 45°, се развлекува страничната капка вода кон средишната. По нивно спојување, покровното стакленце се спушта на предметното стакленце.
