English: Preparing a fixed smear of yeast cells
Procedures: 1. One drop of physiological solution is to be applied on a clean microscopy slide. 2. Using an inoculation loop, inoculum is to be transferred from a plate containing visibly grown yeast colonies. Smearing of the inoculum is to be performed. 3. Heat fixation of the smear is to be performed. 4. Fushcin is to be applied and allowed to dye present cells for 1-2 minutes. 5. The slide is to be washed with water and dried. 6. The slide is to be observed under microscope. RESULTS: The MEA plate should show viable growth of yeast colonies. The microscopyslide should show oval pink eukaryotic cells. Постапки: 1. На чисто предметно стакленце се капнува капка физиолошки раствор. 2. Со еза се зема од плочата со изразнат квасец и се прави размаз на предметно стакленце. 3. Размазот се фиксира на пламен. 4. Се прелива размазот со фуксин и се остава да делува 1-2 минути. 5. Препаратот се измива го со вода и се суши високо над пламен. 6. Препаратот се набљудува прво на објектив со најмало зголемувње, а потоа и на објектив со средно зголемување. РЕЗУЛТАТ: На микроскопскиот препарат се забележуваат овални, розево обоени, со типични еукариотски структури. Осмислено и изведено од Софија Костандиновска, Институт за биологија, ПМФ, УКИМ, Скопје. |
