English: Physical factors of microbial growth using ultraviolet radiation
Procedures: 1. With a sterile inoculation loop, E.coli is to be inoculated onto five Endo agar plates. 2. One plate is to serve as control, one is to be exposed under UV light for 5 sec, one for 15 sec and one for 1 min. 3. The plates are incubated at 37 oC for 24h. 4. The final plate is to be exposed to UV light for 3 min, with the plates lid placed appropriately on the plate. 5. The final plate is to be incubated along with the rest under identical conditions. CAUTION: DO NOT STARE DIRECTLY INTO UV LIGHT! IT CAN DAMAGE EYES! RESULTS: UV has relatively low penetrability and as such can only destroy microorganisms present on surfaces. The lid serves to block UV, and the plate in question should show the same growth as the control. UV light in the range of 260-270 nm possesses the greatest microbiocidal properties as DNA absorbs photons most readily from this range, forming thymine-thymine dimers, halting any replication process. This is the same mechanisms which allows UV to cause somatic mutations elsewhere. Planned and performed by Sofija Kostandinovska, Institute of Biology, FNSM, Ss. Постапки: 1. Со претходно стерилизирана еза, се засејуваат сите 5 плочи од Ендо агар со течна култура од E.coli. 2. Првата плоча е контрола, а втората плоча се изложува на UV светлина 5 секунди, втората 15 секунди, третата 1 минута. 3. Плочите се затвораат со капак и се инкубираат 24 часа, на 37 oC. 4. Петтата плоча, подготвена на истиот начин, но со ставен капак, се изложува на UV светлина 3 минути. 5. Плочата се инкубира заедно со останатите плочи. Предупредување: Не гледајте директно во UV светлината! Може да ги оштети вашите очи! РЕЗУЛТАТ: UV светлината има мала пенетрирачка моќ и ги уништува микроорганизмите кои се присутни на површината на материјалот кој се изложува директно на радијација, додека капакот на плочата претставува еден вид на заштита на колониите на E.coli од радијација. Најголема микробицидна активност покажува UV светлината со бранова должина од 260-270 nm. На оваа бранова должина микробната ДНК ја апсорбира UV светлината, која предизвикува соседните тимински бази во истиот ДНК синџир да се врзат ковалентно, и на тој начин се формираат тимин-тимин димери. Како се реплицира ДНК, нуклеотидите не можат комплементарно да се врзат заради овие димери и на тој начин престанува репликацијата на тој синџир ДНК. Всушност, УВ радијацијата на овој начин предизвикува мутација и доведува до погрешна протеинска синтеза. Осмислено и изведено од Софија Костандиновска, Институт за биологија, ПМФ, УКИМ, Скопје. |
