English: Microscopic morphology as a biosignature (A) Marine microplankton, part of the contents of one dip of a hand net, photographed on board the NOAA Ship Oscar Elton Sette off Kona, September 20, 2006. The image contains diverse planktonic organisms, ranging from photosynthetic cyanobacteria and diatoms to many different types of zooplankton, including both holoplankton (permanent residents of the plankton) and meroplankton (temporary residents of the plankton, e.g., fish eggs, crab larvae, worm larvae) (image courtesy David Liittschwager). (B) Low-magnification image of brine collected from sea ice near Nuuk, Greenland, from a sackhole in spring 2015. On this scale, examples of diatoms can be seen. (C) High-magnification bright-field image of Greenland brine showing numerous prokaryotes. (D) The same image from (C) stained with acridine orange and imaged with epifluorescence microscopy. Because staining labels DNA, this technique permits distinguishing bacteria from debris. The combination of multiple dyes, such as acridine orange and DAPI, can further remove ambiguity caused by nonspecific adherence of dyes to minerals. |
