A Em circunstâncias normais a ApoE forma complexos com Aβ (ApoE/Aβ) e interagindo com o LRP1, promove a sua eliminação do CNS para os capilares, através da BHE. Também através deste receptor em pericitos, inibe a ativação de MMPs, promovendo a integridade da BHE. B A ApoE4 atrasa a eliminação de Aβ através da BHE, deslocando a eliminação do LRP1 para o VLDLR, que possuí uma taxa muito mais lenta de endocitose. Esta apolipoproteína, ao não interagir com o LRP1, também reduz a integridade da BHE através da ativação de MMPs em pericitos, levando à degradação de proteínas de junções aderentes e de oclusão no interior das células endoteliais, perturbando assim a integridade da barreira.
