Description
Deutsch: Nobelpreis für Physiologie oder Medizin 2019: Zelluläre Anpassungsmechanismen an die Verfügbarkeit von Sauerstoff. Bei normaler zellulärer Verfügbarkeit von Sauerstoff wird Hif-1 alpha an zwei Prolin-Resten hydroxyliert. Daraufhin assoziiert es mit VHL und wird mit Ubiquitin markiert. Hif-1 alpha wird dann im Proteasom abgebaut. Bei Sauerstoffmangel hingegen steht kein Sauerstoff zur Verfügung, um die Prolin-Reste zu hydroxylieren. Daraufhin transloziert Hif-1 alpha in den Zellkern, wo es an Hif-1 beta (auch ARNT genannt) assoziiert. Dieser Komplex bindet an die DNA-Region HRF (Hypoxie responsives Element), woraufhin die zugehörigen Gene abgelesen werden. Diese sind an der Regulation einer Vielzahl von Prozessen beteiligt, so der Erythropoese, der Glykolyse und der Angiogenese.
