Description
English: Figure 1 JAK2 inhibition in Graft-versus-Host Disease. Janus kinases are crucial to mediate extracellular signals. Binding of cytokines results in receptor dimerization and phosphorylation, subsequently phosphorylating STAT molecules by receptor tyrosine kinases. Phosphorylation of STAT leads to dimerization and translocation into the nucleus, followed by enhancing gene transcription. JAK/STAT signaling is important in regulating cell activation, proliferation, migration and effector cytokine production, thereby enhancing GvHD severity. JAK1/2 inhibition by ruxolitinib reduces pro-inflammatory signaling and cell migration, resulting in reduced GvHD disease progression. Created with Biorender.com.
