Pollen grains of common pine (Pinus silvestris) stained with acetocarmine

Summary[edit]

Description
English: Determination of pollen viability of Pinus silvestris using the acetocarmine method. For this purpose, pollen is collected and either stained immediately or fixed in Carnua fixative for two hours or more, then pollen grains are transferred into 80% ethyl alcohol, where they are stored. Acetocarmine staining is carried out at any time.

Pollen grains are placed on a slide, a couple of drops of acetocarmine are dropped, covered with a coverslip, and gently heated over a spirit flame. Acetocarmine is a nuclear dye; therefore, in fertile pine pollen grains the granular cytoplasm, the vegetative cell, the antheridial cell, etc. are stained carmine red. Sterile pollen grains are not stained with carmine or are stained irregularly.
