Polski: (a) Zymografia żelowa (IGZ) i (b) zymografia żelowo-transferowa (TGZ) (a) W IGZ substrat jest włączony do żelu SDS-PAGE. Po rozdzieleniu według względnych mas cząsteczkowych żel inkubuje się przy optymalnym pH i temperaturze w celu wywołania reakcji proteolitycznej. Żel jest następnie barwiony, a prążki hydrolityczne są przejrzyste na ciemnym tle. (b) W TGZ próbki najpierw rozdziela się na zwykłym żelu SDS-PAGE. Następnie żel rozdzielający układa się na jednym lub wielu żelach zawierających substrat i białka przenosi się na żele substratu. Te ostatnie są przemywane, inkubowane w celu przeprowadzenia reakcji hydrolitycznej, a następnie barwione. Prążki hydrolityczne są zabarwione. |
