English: Top: schematic representation of the viral vector nucleic acid of valoctocogene roxaparvovec: ITR = inverted terminal repeats (double-stranded); single-stranded: HLP = hybrid liver-specific promoter, LS = leader sequence, hFVIII-SQ = codon-optimized sequence encoding hFVIII-SQ protein, sPA = synthetic polyadenyl sequence.
Bottom: schematic representation of the expressed hFVIII-SQ protein, consisting of the heavy chain (HC) with protein domains A1 and A2 and the light chain (LC) with protein domains A3, C1, and C2. The wild-type B domain between the A2 and A3 domains is replaced by a 14 amino acid “SQ” linker sequence, from the normal B domain sequence (acc. to description in: Assessment Report Roctavian, Committee for Medicinal Products for Human Use (CHMP), 23.
