English: Striatal neuron (trasgenic YAC128 mouse model of Huntington disease) after immunocytochemical staining for DARPP-32 protein.
Antigen-antibody reaction was performed with primary rabbit antibodies on the marker protein of medium-spiny neurons (13 DIV), secondary antibodies to rabbit antibodies fused to the fluorophore (Alexa Fluor 488). The image was made with a laser confocal scanning microscope Zeiss LSM 700 .Русский: Среднешипиковый нейрон стриатума (трансгенная линия мышей YAC 128) после иммуно-цитохимического окрашивания на маркерный белок DARPP-32.
Реакция антиген-антитело произведена c помощью первичных кроличьих антител на маркерный белок средне-шипиковых нейронов (13 дней развития in vitro), вторичные антитела - к антителам кролика, слитые с флуорофором (Alexa Fluor 488). Снимок получен с помощью лазерного конфокального сканирующего микроскопа Zeiss LSM 700.English: For the preparation of cultures, the brain of newborn transgenic mice (1-2 days) of the YAC128 line (Jackson Labs, 004938), which is the carrier of the human mutant HTT allele, was used.The mice were genotyped by PCR using primers to 44 and 45 exons of the human HTT gene, after which the brain was removed and the primary striatum culture was made. The cells were cultured on 12 mm glass coated poly-L-lysine (Sigma) in Neurobasal-A (Invitrogen) supplemented with B-27 (Gibco), FBS (Gibco) and L-glutamine (Invitrogen). The cultures were incubated for 13 days at 37 ° C and 5% CO2. |
