The column is connected to an Äkta Avant, which has a refrigerated fraction collector, but most of the columns designed for it are not the cooling (jacketed) type. This is a low tech solution, which we used when loading large sample volumes directly to the column. Loading large sample volumes takes a long time and we do not want the protein to be at room temperature for many hours. When the bottle is filled with ice water, it keeps the column cold for about 3-6 hours (depending on the ambient temperature).
