Dividing 3T3 cells in 1.38 squre mm area of slide, stained by DAPI, fixed with 1% Glutar aldehide MAX MI DAPI 9-07-2015, A2 well of 24 well plate. Acquired at Yokogawa CSU-X1 confocal scaning head Objective x63 oil, Rolera EM CCD 1024x1024 Image Processing done ZEN 2012 (Carl Zeiss) Exported to TIFF as one image of 10x10 adjacent fields of view (FOV), 9kx9k pxls, cuttoff piece at 2x2 FOV FIJI/ImageJ (NIH openware) Z projection of 5 focal planes, "Rainbow" LUT, brighteness and contrast adjustion. |
