Description
English: Supplementary Movie 4 A-lattice enriched MT seeds (green) were co-assembled from pig brain tubulin labelled with Alexa-488, Mal3FL and GMPCPP. Seeds were attached to flow cells coated with anti-Alexa488 antibodies. Flow cells were flushed with buffer to remove Mal3 and unbound MTs. Buffer containing GTP and unlabelled S. pombe single isoform tubulin (grey) was added and MT dynamics observed using timelapse epifluorescence (for seeds) and darkfield (for dynamic unlabelled S. pombe MTs) microscopy. MTs grown from the A-lattice enriched seeds had frequent episodes of transient short MT growth, which we have named piku-piku or MT dithering (blue label). Time is in minutes. Scale bar is 5 μm.
