J. Mann, Chemical Aspects of Biosynthesis, Oxford University Press, pp.42-43 (1994) ISBN 0-19-855676-4.の説明図に酵素・補酵素を加筆改変。赤字は反応部位。この反応機構はE.J.コーリーにより立体構造とともに提唱された。その後の機器分析により立体構造は正しいことは確認されている。

解説

スクアレンエポキシダーゼによりスクアレンの2,3位がエポキシ化すると、安定配座のひとつであるchar-boat-char-boat conformationより、自発的に閉環反応と転移反応が協奏的に進行しラノステロールが進行する。別の安定配座のchar-char-char-boat conformationからは5つシクロヘキサン環が縮合したテルペノイドのβamyrinなどが生成する。ラノステロールは還元と酸化的脱メチル化を繰り返しコレステロールへと導かれる。
